This year, we advanced understanding of melanoma tumor biology by demonstrating that UVB cooperates with BRAFV600E and LKB1 loss to impair DNA damage repair and drive a neural-like melanoma phenotype. We also showed that neonatal UVB exposure induces transcriptional reprogramming that bypasses BRAFV600E-driven growth arrest, enabling melanoma initiation independently of nevus progression. Several UV-regulated genes were linked to poor survival, enhanced transformation, and multidrug resistance, emerging as potential therapeutic targets, including UPP1. Clinically, we found that the mutational landscape of SPTCL is broader than previously recognized, identifying TP53 P72R and DNMT3A T735C as relevant contributors, while LEP lacks these alterations, supporting distinct pathogenic mechanisms. We also documented severe vitiligo-like depigmentation following imiquimod treatment for superficial BCC, with recurrence despite intense inflammation.
